What it actually does
Selank begins with the four residues of tuftsin (Thr-Lys-Pro-Arg) and ends in a Pro-Gly-Pro extension at the C-terminus, the same stabilising motif used in Semax.
Research interest centres on GABAergic signalling, the brain’s main inhibitory neurotransmitter system, and on anxiolytic-like effects observed in preclinical models.
Selank has also been studied for its effects on enzymes that break down enkephalins, the body’s own opioid peptides.
The science in detail
Tuftsin is a naturally occurring tetrapeptide (Thr-Lys-Pro-Arg) released from the heavy chain of immunoglobulin G, and is known for immunomodulatory activity. Selank was developed in Russia by extending this sequence with a C-terminal Pro-Gly-Pro motif, giving the heptapeptide Thr-Lys-Pro-Arg-Pro-Gly-Pro.
The Pro-Gly-Pro extension is intended to improve stability: short peptides such as tuftsin are rapidly degraded by peptidases, and the added tail slows that breakdown in experimental systems.
Mechanistic studies have focused on GABAergic signalling. Work in rodents has reported changes in the expression of genes involved in GABAergic neurotransmission, and Selank has been examined for anxiolytic-like effects in preclinical behavioural models.
A separate line of research concerns enkephalin-degrading enzymes. Selank has been reported to inhibit enzymes that break down enkephalins, which would be expected to prolong the activity of these endogenous opioid peptides. As with Semax, much of the literature comes from Russian research groups.
